P09.03.A Associations of levetiracetam use with the safety and tolerability of chemoradiotherapy for patients with newly diagnosed glioblastoma

Abstract Background Levetiracetam (LEV) is one of the most frequently used antiepileptic drugs (AED) for brain tumor patients with seizures. We hypothesized that toxicity LEV and temozolomide-based chemoradiotherapy may overlap. Patients Methods In a retrospective analysis individual patient data using pooled cohort newly diagnosed glioblastoma included in clinical trials prior to (CENTRIC, CORE, AVAglio) or maintenance therapy (ACT-IV), we tested associations hematologic toxicity, nausea emesis, fatigue, psychiatric adverse events during concomitant treatment use alone other AED versus combination no at start treatment. Results Of 1681 2020 who started temozolomide, respectively, 473 714 (28.1% 35.3%) were treated LEV-containing regimen, 538 475 (32.0% 23.5%) AED, 670 831 (39.9% 41.1%) had AED. was associated higher risk univariable multivariable analyses (RR 1.86 1.88, p<0.001) while there fatigue. reduced emesis (HR=0.80, p=0.017) events. Conclusion Any association did not persist beyond phase. Antiemetic properties be beneficial temozolomide.
